Mechanism

• Increases lysosomal pH in antigen presenting cells

• Blocks the activation of toll-like receptors on plasmacytoid dendritic cells (PDCs)

  • Toll-like receptor 9 (TLR 9) recognizes DNA-containing immune complexes

  • => production of interferon

  • => dendritic cell maturation and presentation of antigen to T cells

• Hydroxychloroquine thus reduces the activation of dendritic cells thus mitigating the inflammatory process
